Study on the Breeding and Characterization of High-Efficiency Oil-Degrading Bacteria by Mutagenesis

In the present study, a high-efficiency petroleum hydrocarbon-degrading bacterium MX1 was screened from petrochemical wastewater sludge, and identified using morphological, physiological, biochemical experiments combined with 16S rDNA. Results showed that strain belongs to Enterobacter sp. The degradation conditions were an incubation time of 18 days, temperature 30 °C, pH 7, salinity 2% (w/v), proportion 37.41% for 7 days. combination microwave ultraviolet mutagenesis yielded MXM3U2. mutant had hydrocarbon breakdown efficiency 56.74% after days culture, this value 51.66% higher than original strain. number strains rate n-alkanes (C16, C24, C32, C40) decreased steadily increase in carbon chains test. GC/MS (Gas chromatography mass spectrometry) results process degrading crude oil, hydrocarbons C < 24 degraded first, followed by > 24. good effect on pristane, naphthalene, phenanthrene. via microwave-ultraviolet composite technology.
